Running too far, too fast, and too long speeds progress 'to finish line of life'

ScienceDaily (Nov. 29, 2012) ? Vigorous exercise is good for health, but only if it's limited to a maximum daily dose of between 30 and 50 minutes, say researchers in an editorial published online in Heart.

The idea that more and more high intensity exercise, such as marathons, can only do you good, is a myth say the US cardiologists, and the evidence shows that it's likely to more harm than good to your heart.

"If you really want to do a marathon or full distance triathlon, etc, it may be best to do just one or a few and then proceed to safer and healthier exercise patterns," they warn.

"A routine of moderate physical activity will add life to your years as well as years to your life. In contrast, running too far, too fast, and for too many years may speed one's progress to towards the finishing line of life."

Mitch Ditkoff: Flowers First, Business Second

The other day a sudden fit of gratitude, I bought a dozen roses and brought them home to my wife.

Usually, when I think of buying roses, I go through a predictable sequence of events. First, I surrender to a feeling of expansiveness that takes me over. Then I get curious and smell the flowers. Then I ask the shopkeeper how long she thinks the roses will last.

This is following by calculating the per stem price, doing the math and reaching the pitifully male conclusion that $46.95 is way too much too spend on something that won't last the week and is probably much less expensive somewhere else and it's obviously indulgent of me to be buying so many roses when I've got two kids to put through college and besides, beauty is within.

All of this, of course, is my inner Woody Allen taking the low road in response to what is obviously a Johnny Depp moment.

So I dig deep and bring the roses home -- my entire living room taking shape around them. I become immediately aware of a trend even Republican pollsters could not deny -- there are definitely not enough flowers in the living room. Somehow, the recent appearance of roses has made the rest of the room painfully barren. Tabletops and shelves that only minutes ago were doing just fine, are now utterly flowerless.

So I do the only thing a man can do when faced with such a situation -- I return to the flower shop.

But the shop is closed. Closed? Impossible! I need flowers! So I get back into my car and speed my way to the other flower shop in town. It, too, is closed -- or, should I say, closing. The owner is shutting the door and giving me the "too-bad-you-didn't-get-here a few-minutes-ago" look.

But I will not be denied. And he knows it.

"What do you want?" he asks.

"Flowers," I reply.

He signals me to enter and I buy way more flowers than makes sense. A ridiculous amount.

Let's put it this way: if I was in the federal witness protection program, my sudden flower buying behavior would have made my government handlers very nervous.

Fast forward ten minutes to my wife in our kitchen.

She is looking at me as if I am totally insane -- me, the guy who, only yesterday was making an airtight case for a more modest household budget.

Yes, I am insane, but in a good way. Here's my philosophy: Flowers first. Business second. If money is tight, buy more flowers. The more flowers you buy, the more money will appear. And if not in this lifetime, then the next (or maybe the one after that).

OK. There you go -- my not very financially sound, flower-centric view of the universe. Not quite as newsworthy as credit default swaps, but a whole lot more dependable.

Radio's Laura Ingraham off the air

NEW YORK (AP) ? Radio talk show host Laura Ingraham is off the air ? at least temporarily.

She said Tuesday that she's leaving her distributor, Talk Radio Network, after more than nine years because her contract is up. Ingraham said she is in discussions with other distributors about her show, which currently airs on 325 stations nationally.

Talkers magazine says the conservative Ingraham is the most-listened to woman on radio, with an estimated 5.75 million listeners a week. Talkers said that ranks her among the top eight radio hosts overall.

In the meantime, she said she'll keep her opinions flowing on her website, on Twitter and during Fox News Channel appearances.

The Folly of my Youth and the Mid Life Crisis: Death Sucks

My father in law is dying. ?I know I have mentioned it before but now it is getting serious. ?After a year and a half of threatening to die I think he actually might go and do it in the next few days. ?And I am exhausted.

Why am I exhausted? ?If you are family and reading this you might be really thinking that. ?I mean I haven't been sitting at the house for 8 hours getting up and down with him and mom, I haven't taken any night shifts, I wasn't there last night for all the crying out in pain and emotional turmoil as they waited for someone to deliver the morphine and the wait for it to take affect.

I ?have done nothing.?Literally. I have done nothing for the past few days. ?I am just standing at the ready. ?Afraid to involve myself in any project, just sitting and waiting. ?A million things on my to do list, my son home from college and yet here I am. ?Waiting. ?My job is support. ?I mainly support my husband but try to have a loving word or scripture for the family as well. ?But mostly I am just waiting for John to come home and talk to me, tell me the latest, and experience the emotions. ?Normally I get to break down but this time it is his turn and I will be the rock. ?And it is exhausting and I don't want to do anything. ?I don't even want to write this but I am trying to force myself to do something this afternoon and I usually find it helpful to write out my feelings. ?So I am sharing with you that I would rather be doing something destructive. ?Eating everything in sight or, better yet, a little retail therapy, buy everything in sight!

So now that you know I am totally not feeling it right now I am none-the-less going to share a few scriptures that I am finding encouraging. ?I ?have brief moments where I decide not to run away from my emotions and here are a few things I have found during those times:

Ps 100:4-5 "Enter his gates with thanksgiving and his courts with praise; give thanks to him and praise his name. ?For the Lord is good and is love endures forever; his faithfulness continues through all generations."

I am calling that one the verse that starts off for Don and ends for the rest of us. ?Don is going to enter the Gates of heaven but his faithfulness, Don's steadfast and unwavering devotion to the Lord, continues through the generations that come behind him. ?The generations that he has influenced. ?And while we are quick to think of his family Don has made a difference in the lives of men and women well beyond his family through his teaching both of the Word at church and through his teaching of Advanced First Aid (now EMT) at Normandale. ?Many a Minneapolis cop got their initial first aid training from John's parents in the last 80's and early 90's. ?How thankful we all are, how blessed and rich our lives have been because of the gift of Don.

I read other things and think of Don:

Is 21:3-4 "At this my body is racked with pain, pangs seize me, like those of a woman in labor; I am staggered by what I hear and bewildered by what I see. ?My heart falters, fear makes me tremble; the twilight I longed for has become a horror to me."

I know, that isn't encouraging at all. ?But I love how the bible can express pain as well as give hope. ?I don't know why some people have to die so painfully. ?It doesn't seem fair. ?This is not a man who deserves this death. ?Even yesterday when I was over there we were laughing as he started asking John's mom if she knew the Truth. ?As his mind goes what is left is the Truth of God's love and his continued need to share it with others. ?John drove their car home last night as Jake and I both needed cars yesterday. ?On the floor of the passenger side are the scripture memory cards his parents continue to read and memorize. When I mentioned Ps 100:4-5 at the house John went to get the bible to read it. ?John's mom kept saying, "recite it". ?I bet she already knew what he was going to read. ?God doesn't promise us freedom from pain or trials in this life on earth but he does promise us that freedom in heaven and soon Don will follow Jesus through those gates and into that peace.

And finally, and again this isn't really for encouragement here just something I read and thought about,

Proverbs 25:20 "Like one who takes away a garment on a cold day, or like vinegar poured on soda, is one who sings songs to a heavy heart."

When someone is struggling, it isn't helpful to be cheerful. ?This sucks! ?Watching this man die sucks. ?Even if this wasn't a hard and long death, if he has simply died peacefully in his sleep, this would suck. ?I will not be denied the right to grieve nor will I deny my family that right. ?We are not alone, God is with us, and one day we will sing songs again but today we will feel and experience these moments of pain and grief and be stressed and overwhelmed.

Bleh...

Off to switch the laundry. ?I like to be prepared with clean underwear in all situations.

60 percent of lower 48 states now in drought

ST. LOUIS -- The worst U.S. drought in decades has deepened again after more than a month of encouraging reports of slowly improving conditions, a drought-tracking consortium said Wednesday, as scientists struggled for an explanation other than a simple lack of rain.

While more than half of the continental U.S. has been in a drought since summer, rain storms had appeared to be easing the situation week by week since late September. But that promising run ended with Wednesday's weekly U.S. Drought Monitor report, which showed increases in the portion of the country in drought and the severity of it.

The report showed that 60.1 percent of the lower 48 states were in some form of drought as of Tuesday, up from 58.8 percent the previous week. The amount of land in extreme or exceptional drought ? the two worst classifications ? increased from 18.3 percent to 19.04 percent.

Grain prices soar as drought impact deepens

The Drought Monitor's map tells the story, with dark red blotches covering the center of the nation and portions of Texas and the Southeast as an indication of where conditions are the most intense. Those areas are surrounded by others in lesser stages of drought, with only the Northwest, Florida and a narrow band from New England south to Mississippi escaping.

The governor of Missouri has enacted an emergency measure to drill new wells in areas where water is scarce, providing much-needed relief for the state's farmers and ranchers. NBC's Thanh Truong reports.

A federal meteorologist cautioned that Wednesday's numbers shouldn't be alarming, saying that while drought usually subsides heading into winter, the Drought Monitor report merely reflects a week without rain in a large chunk of the country.

"The places that are getting precipitation, like the Pacific Northwest, are not in drought, while areas that need the rainfall to end the drought aren't getting it," added Richard Heim, a meteorologist with the National Oceanic and Atmospheric Administration's National Climatic Data Center. "I would expect the drought area to expand again" by next week since little rain is forecast in the Midwest in coming days.

Drought conditions plague much of the United States after a summer of scorching temperatures and a lack of rain. The dryness is affecting America's farmland, threatening crops like soybean and corn.

He said there was no clear, scientific explanation for why the drought was lingering or estimate for how long it would last.

"What's driving the weather? It's kind of a car with no one at the steering wheel," Heim said. "None of the atmospheric indicators are really strong. A lot of them are tickling around the edges and fighting about who wants to be king of the hill, but none of them are dominant."

White House offers drought relief, feels heat to waive ethanol mandate

As the drought continues, ranchers worry for the future especially now that the total number of cattle in the U.S. is already the smallest in 60 years. NBC's Kristen Dahlgren reports.

The biggest area of exceptional drought, the most severe of the five categories listed by the Drought Monitor, centers over the Great Plains. Virtually all of Nebraska is in a deep drought, with more than three-fourths in the worst stage. But Nebraska, along with the Dakotas to the north, could still see things get worse "in the near future," the USDA's Eric Luebehusen wrote in Wednesday's update.

The drought also has been intensifying in Kansas, the top U.S. producer of winter wheat. It also is entirely covered by drought, and the area in the worst stage rose nearly 4 percentage points to 34.5 percent as of Tuesday. Much of that increase was in southern Kansas, where rainfall has been 25 percent of normal over the past half year.

Peggy Ebbesmeyer's ranch in Truxton, Missouri has been hit hard by drought.

After a summer in which farmers watched helpless as their corn dried up in the heat and their soybeans became stunted, many are now worrying about their winter wheat.

It has come up at a rate on par with non-drought years, but the quality of the drop doesn't look good, according to the USDA. Nearly one-quarter of the winter wheat that germinated is in poor or very poor condition, an increase of 2 percentage points from the previous week and 9 percentage points worse than the same time in 2011. Forty-two percent of the plantings are described as in fair shape, the same as last week.

Farmers who might normally irrigate in such circumstances worry about low water levels in the rivers and reservoirs they use, and many are hoping for snow to ease the situation. But it would take a lot. About 20 inches of snow equals just an inch of actual water, and many areas have rain deficits of a foot or more.

Hurricane Sandy Relief Purpose

This announcement provides relief to taxpayers who have been adversely affected by Hurricane Sandy and have retirement assets in qualified employer plans they would like to use to alleviate hardships caused by Hurricane Sandy. In addition, this announcement provides relief from certain verification procedures that may be required under retirement plans with respect to loans and hardship distributions. The relief provided under this announcement is in addition to the relief already provided by the Service pursuant to News Release IR-2012-83 under ?7508A of the Internal Revenue Code (?Code?) for victims of Hurricane Sandy. (See the regulations under ?7508A and Section 8 of Rev. Proc. 2007-56, 2007-34 I.R.B. 388, for a listing of employee benefit-related acts currently postponed until February 1, 2013, because of the disaster.)

Background

The laws relating to qualified employer plans impose various limitations on the permissibility of loans and distributions from those plans. For example, ?401(k)(2)(B)(i) of the Code provides that in the case of a ?401(k) plan that is part of a profit-sharing or stock bonus plan, elective deferrals may be distributed only in certain situations, one of which is on account of hardship. Section 403(b)(11) provides similar rules with respect to elective deferrals under a ?403(b) plan. Section 457(d)(1)(A) provides that a plan described in ?457(b) may not permit distributions before the occurrence of certain enumerated events, one being when the participant is faced with an unforeseeable emergency. Certain other types of plans or accounts are not permitted to make in-service distributions (distributions to a participant who is still an employee) even if there is a hardship. For example, in-service hardship distributions are generally not permitted from pension plans or from accounts holding qualified nonelective contributions (?QNECs?) described in ?401(m)(4)(C) or qualified matching contributions (?QMACs?) described in ?401(k)(3)(D)(ii)(I). However, Rev. Rul. 2004-12, 2004-7 I.R.B. 478, holds that if amounts attributable to rollover contributions are separately accounted for within a plan, those amounts may be distributed at any time, pursuant to the employee's request. Section 72(p) imposes certain requirements relating to plan loans. Unless those requirements are satisfied, a loan is treated as a distribution under the plan.

In order to make a loan or distribution (including a hardship distribution), a plan must contain language authorizing the loan or distribution. Also, except to the extent a distribution consists of already-taxed amounts, the distribution will be includible in gross income and generally subject to the 10-percent additional tax under ?72(t). Similar rules relating to income inclusion and taxation apply to a distribution from an IRA.

Plan provisions and regulations under certain Code sections establish verification procedures that a plan must follow before loans or distributions can be made from the plan. For example, the regulations under ?401(k) set forth certain criteria an employee must meet in order to receive a hardship distribution. A plan may contain procedures designed to confirm that the criteria have been satisfied.

Relief

As described below, a qualified employer plan will not be treated as failing to satisfy any requirement under the Code or regulations merely because the plan makes a loan, or a hardship distribution for a need arising from Hurricane Sandy, to an employee or former employee whose principal residence on October 26, 2012, was located in one of the counties or Tribal Nations that have been identified as covered disaster areas because of the devastation caused by Hurricane Sandy or whose place of employment was located in one of these counties or Tribal Nations on that date or whose lineal ascendant or descendant, dependent or spouse had a principal residence or place of employment in one of these counties or Tribal Nations on that date. Covered disaster areas are identified as federally declared disaster areas in the News Releases issued by the IRS for Victims of Hurricane Sandy, which are found on IRS.gov at:? http://www.irs.gov/uac/Newsroo.... Plan administrators may rely upon representations from the employee or former employee as to the need for and amount of a hardship distribution, unless the plan administrator has actual knowledge to the contrary, and the distribution is treated as a hardship distribution for all purposes under the Code and regulations.

For purposes of this announcement, a ?qualified employer plan? means a plan or contract meeting the requirements of ?401(a), 403(a) or 403(b), and, for purposes of the hardship relief, which could, if it contained enabling language, make hardship distributions. For purposes of this paragraph, a ?qualified employer plan? also means a plan described in ?457(b) maintained by an eligible employer described in ?457(e)(1)(A), and any hardship arising from Hurricane Sandy is treated as an ?unforeseeable emergency? for purposes of distributions from such plans. For example, a profit-sharing or stock bonus plan that currently does not provide for hardship or other in-service distributions may nevertheless make Sandy-related hardship distributions pursuant to this announcement, except from QNEC or QMAC accounts or from earnings on elective contributions (see below for plan amendment requirements). A defined benefit or money purchase plan, which generally cannot make in-service hardship distributions, may not make hardship distributions pursuant to this announcement, other than from a separate account, if any, within the plan containing either employee contributions or rollover amounts.

The amount available for hardship distribution is limited to the maximum amount that would be permitted to be available for a hardship distribution under the plan under the Code and regulations. However, the relief provided by this announcement applies to any hardship of the employee, not just the types enumerated in the regulations, and no post-distribution contribution restrictions are required. For example, regulations under ?401(k) provide safe harbor hardship distribution standards under which a hardship is deemed to exist only for certain enumerated events, and after receipt of the hardship amount, the employee is prohibited from making contributions for at least 6 months. Plans need not follow these rules with respect to hardship distributions for which relief is provided under this announcement.

To make a loan or hardship distribution, a qualified employer plan that does not provide for them must be amended to provide for loans or hardship distributions no later than the end of the first plan year beginning after December 31, 2012. To qualify for the relief under this announcement, a hardship distribution must be made on account of a hardship resulting from Hurricane Sandy and be made on or after October 26, 2012, and no later than February 1, 2013. Plan loans made pursuant to this announcement must satisfy the requirements of ?72(p).

In addition, a retirement plan will not be treated as failing to follow procedural requirements for plan loans (in the case of retirement plans other than IRAs) or distributions (in the case of all retirement plans, including IRAs) imposed by the terms of the plan merely because those requirements are disregarded for any period beginning on or after October 26, 2012, and continuing through February 1, 2013, with respect to distributions to individuals described in the first paragraph under ?Relief?, above, provided the plan administrator (or financial institution in the case of distributions from IRAs) makes a good-faith diligent effort under the circumstances to comply with those requirements. However, as soon as practicable, the plan administrator (or financial institution in the case of IRAs) must make a reasonable attempt to assemble any forgone documentation. For example, if spousal consent is required for a plan loan or distribution and the plan terms require production of a death certificate if the employee claims his or her spouse is deceased, the plan will not be disqualified for failure to operate in accordance with its terms if it makes a loan or distribution to an individual described in the first paragraph under ?Relief? in the absence of a death certificate if it is reasonable to believe, under the circumstances, that the spouse is deceased, the loan or distribution is made no later than February 1, 2013, and the plan administrator makes reasonable efforts to obtain the death certificate as soon as practicable. Taxpayers are reminded that in general the normal spousal consent rules continue to apply. For purposes of this announcement, ?retirement plan? has the same meaning as ?eligible retirement plan? under ?402(c)(8)(B).

Israel says prefers diplomacy over Gaza invasion option

GAZA/JERUSALEM (Reuters) - Israel bombed dozens of targets in Gaza on Monday and said that while it was prepared to step up its offensive by sending in troops, it preferred a diplomatic solution that would end Palestinian rocket fire from the enclave.

As international pressure mounted for a truce, mediator Egypt said a deal to end the fighting could be close.

Twelve Palestinian civilians and four fighters were killed in the air strikes, bringing the Gaza death toll since fighting began on Wednesday to 90, more than half of them non-combatants, local officials said. Three Israeli civilians have been killed.

After an overnight lull, militants in the Hamas-run Gaza Strip fired 45 rockets at southern Israel, causing no casualties, police said. One damaged a school, but it was closed at the time.

The deaths of 11 Palestinian civilians - nine from one family - in an air strike on Sunday - drew more international calls for an end to six days of hostilities and could test Western support for an offensive Israel billed as self-defense after years of cross-border rocket attacks.

Israel's military did not immediately comment on a report in the liberal Haaretz newspaper that it had mistakenly fired on the Dalu family home, where the dead spanned four generations, while trying to kill a Hamas rocketry chief.

Echoes of explosions in Gaza mixed with cries of grief and defiant chants of "God is greatest" at the funeral of the four children and five women killed in the attack that flattened the three-storey house. Their bodies were wrapped in Palestinian and Hamas flags and thousands turned out to mourn them.

United Nations Secretary-General Ban Ki-moon was due to arrive in Cairo to weigh in on ceasefire efforts led by Egypt, which borders both Israel and Gaza and whose Muslim Brotherhood-rooted government has been hosting leaders of Hamas and Islamic Jihad, a smaller armed faction in the Palestinian enclave.

Israeli media said a delegation from Israel had also been to Cairo for the truce talks. A spokesman for Prime Minister Benjamin Netanyahu's government declined comment on the matter.

"Israel is prepared and has taken steps, and is ready for a ground incursion which will deal severely with the Hamas military machine," a senior official close to Netanyahu told Reuters.

But he added: "We would prefer to see a diplomatic solution that would guarantee the peace for Israel's population in the south. If that is possible, then a ground operation would no longer be required. If diplomacy fails, we may well have no alternative but to send in ground forces."

The official's language echoed that of U.S. President Barack Obama, who said on Sunday it would be "preferable" to avoid a move into Gaza. Obama also said Israel had a right to self-defense and no country would tolerate missiles raining down on its citizens.

Egyptian negotiators could be close to achieving a deal between Israel and the Palestinians to stop the fighting could be close, the Egyptian prime minister said.

"I think we are close, but the nature of this kind of negotiation, (means) it is very difficult to predict," Hisham Kandil said in an interview in Cairo for the Reuters Middle East Investment Summit.

Egypt's foreign minister is expected to visit Gaza on Tuesday with a delegation of Arab ministers to express solidarity with the Palestinians.

In scenes recalling Israel's 2008-2009 winter invasion of Gaza, tanks, artillery and infantry have massed in field encampments along the sandy, fenced-off Gaza border and military convoys moved on roads in the area.

Israel has also authorized the call-up of 75,000 military reservists, so far mobilizing around half that number.

WORLD CONCERN

The Gaza fighting has stoked the worries of world powers watching an already combustible region.

In the absence of any prospect of permanent peace between Israel and Hamas and other Islamist factions, mediated deals for each to hold fire unilaterally have been the only formula for stemming bloodshed in the past. But both sides now placed the onus on the other.

Izzat Risheq, aide to Hamas politburo chief Khaled Meshaal, wrote on Facebook that Hamas would enter a truce only after Israel "stops its aggression, ends its policy of targeted assassinations and lifts the blockade of Gaza".

Listing Israel's terms, Vice Prime Minister Moshe Yaalon wrote on Twitter: "If there is quiet in the south and no rockets and missiles are fired at Israel's citizens, nor terrorist attacks engineered from the Gaza Strip, we will not attack."

Yaalon also said Israel wanted an end to Gaza guerrilla activity in the neighboring Egyptian Sinai, a desert peninsula where lawlessness has spread during Cairo's political crises.

Israel bombed some 80 sites in Gaza overnight, the military said, adding in a statement that targets included "underground rocket launching sites, terror tunnels and training bases" as well as "buildings owned by senior terrorist operatives".

Netanyahu has said he had assured world leaders that Israel was doing its utmost to avoid causing civilian casualties in Gaza. At least 22 of the Gaza fatalities have been children, medical officials said.

Before leaving for Cairo, Ban urged Israel and the Palestinians to cooperate with all Egyptian-led efforts to reach an immediate ceasefire.

But a big rocket strike might be enough for Netanyahu to give a green light for a Gaza invasion, despite the political risks of heavy casualties before a January election he is favored to win.

Although 84 percent of Israelis supported the current Gaza assault, according to a Haaretz poll, only 30 percent wanted an invasion. Nineteen percent wanted their government to work on securing a truce soon.

Israel's declared goal is to deplete Gaza arsenals and force Hamas to stop rocket fire that has bedevilled Israeli border towns for years.

The rockets now have greater range, becoming a strategic weapon for Gaza's otherwise massively outgunned militants. Several projectiles have targeted Tel Aviv and Jerusalem. None hit the two cities and some of the rockets were shot down by Israel's Iron Dome interceptor system.

As a precaution against the rocket interceptions endangering nearby Ben-Gurion International Airport, civil aviation authorities said on Monday new flight paths were being used.

There was no indication takeoffs and landings at Ben-Gurion had been affected.

Hamas and other groups in Gaza are sworn enemies of the Jewish state which they refuse to recognize and seek to eradicate, claiming all Israeli territory as rightfully theirs.

Hamas won legislative elections in the Palestinian Territories in 2006 but a year later, after the collapse of a unity government under President Mahmoud Abbas the Islamist group seized control of Gaza in a brief and bloody civil war with forces loyal to Abbas.

Breakthrough nanoparticle halts multiple sclerosis in mice, offers hope for other immune-related diseases

ScienceDaily (Nov. 18, 2012) ? In a breakthrough for nanotechnology and multiple sclerosis, a biodegradable nanoparticle turns out to be the perfect vehicle to stealthily deliver an antigen that tricks the immune system into stopping its attack on myelin and halt a model of relapsing remitting multiple sclerosis (MS) in mice, according to new Northwestern Medicine research.

The new nanotechnology also can be applied to a variety of immune-mediated diseases including Type 1 diabetes, food allergies and airway allergies such as asthma.

In MS, the immune system attacks the myelin membrane that insulates nerves cells in the brain, spinal cord and optic nerve. When the insulation is destroyed, electrical signals can't be effectively conducted, resulting in symptoms that range from mild limb numbness to paralysis or blindness. About 80 percent of MS patients are diagnosed with the relapsing remitting form of the disease.

The Northwestern nanotechnology does not suppress the entire immune system as do current therapies for MS, which make patients more susceptible to everyday infections and higher rates of cancer. Rather, when the nanoparticles are attached to myelin antigens and injected into the mice, the immune system is reset to normal. The immune system stops recognizing myelin as an alien invader and halts its attack on it.

"This is a highly significant breakthrough in translational immunotherapy," said Stephen Miller, a corresponding author of the study and the Judy Gugenheim Research Professor of Microbiology-Immunology at Northwestern University Feinberg School of Medicine. "The beauty of this new technology is it can be used in many immune-related diseases. We simply change the antigen that's delivered."

"The holy grail is to develop a therapy that is specific to the pathological immune response, in this case the body attacking myelin," Miller added. "Our approach resets the immune system so it no longer attacks myelin but leaves the function of the normal immune system intact."

The nanoparticle, made from an easily produced and already FDA-approved substance, was developed by Lonnie Shea, professor of chemical and biological engineering at Northwestern's McCormick School of Engineering and Applied Science.

"This is a major breakthrough in nanotechnology, showing you can use it to regulate the immune system," said Shea, also a corresponding author. The paper will be published Nov. 18 in the journal Nature Biotechnology.

Miller and Shea are also members of the Robert H. Lurie Comprehensive Cancer Center of Northwestern University. In addition, Shea is a member of the Institute for BioNanotechnology in Medicine and the Chemistry of Life Processes Institute.

Clinical Trial for Ms Tests Same Approach -- With Key Difference

The study's method is the same approach now being tested in multiple sclerosis patients in a phase I/II clinical trial -- with one key difference. The trial uses a patient's own white blood cells -- a costly and labor intensive procedure -- to deliver the antigen. The purpose of the new study was to see if nanoparticles could be as effective as the white blood cells as delivery vehicles. They were.

The Big Nanoparticle Advantage for Immunotherapy

Nanoparticles have many advantages; they can be readily produced in a laboratory and standardized for manufacturing. They would make the potential therapy cheaper and more accessible to a general population. In addition, these nanoparticles are made of a polymer called Poly(lactide-co-glycolide) (PLG), which consists of lactic acid and glycolic acid, both natural metabolites in the human body. PLG is most commonly used for biodegradable sutures.

The fact that PLG is already FDA approved for other applications should facilitate translating the research to patients, Shea noted. Miller and Shea tested nanoparticles of various sizes and discovered that 500 nanometers was most effective at modulating the immune response.

"We administered these particles to animals who have a disease very similar to relapsing remitting multiple sclerosis and stopped it in its tracks," Miller said. "We prevented any future relapses for up to 100 days, which is the equivalent of several years in the life of an MS patient."

Shea and Miller also are currently testing the nanoparticles to treat Type one diabetes and airway diseases such as asthma.

Nanoparticles Fool Immune System

In the study, researchers attached myelin antigens to the nanoparticles and injected them intravenously into the mice. The particles entered the spleen, which filters the blood and helps the body dispose of aging and dying blood cells. There, the particles were engulfed by macrophages, a type of immune cell, which then displayed the antigens on their cell surface. The immune system viewed the nanoparticles as ordinary dying blood cells and nothing to be concerned about. This created immune tolerance to the antigen by directly inhibiting the activity of myelin responsive T cells and by increasing the numbers of regulatory T cells which further calmed the autoimmune response.

"The key here is that this antigen/particle-based approach to induction of tolerance is selective and targeted. Unlike generalized immunosuppression, which is the current therapy used for autoimmune diseases, this new process does not shut down the whole immune system," said Christine Kelley, National Institute of Biomedical Imaging and Bioengineering director of the division of Discovery Science and Technology at the National Institutes of Health, which supported the research. "This collaborative effort between expertise in immunology and bioengineering is a terrific example of the tremendous advances that can be made with scientifically convergent approaches to biomedical problems."

"The idea to couple antigens to nanoparticles was conceived in discussions between Dr. Miller's laboratory, the Myelin Repair Foundation's drug discovery advisory board and Dr. Michael Pleiss, a member of the Myelin Repair Foundation's internal research team, and we combined our efforts to focus on patient-oriented, clinically relevant research with broad implications for all autoimmune diseases. Our unique research model is designed to foster and extract the innovation from the academic science that we fund and transition these technologies to commercialization. The overarching goal is to ensure this important therapeutic pathway has its best chance to reach patients, with MS and all autoimmune diseases."

Searchers in the Gulf of Mexico say they've found the body of one of the two people who went missing after an oil platform explosion on Friday. NBC's Lester Holt reports.?

Updated at 5:40 a.m. ET: Divers found a body Saturday evening below the oil platform that caught fire after an explosion in the Gulf of Mexico, the Coast Guard and an official of the company that owns the rig said.

Black Elk Energy President and CEO John Hoffman told reporters in Houston that a body was spotted on the sea floor by a dive team hired by the company to supplement the Coast Guard search for two workers missing after an explosion and fire wracked the rig on Friday.

"Divers will continue to search for the second missing worker," Hoffman wrote in an email. "Our thoughts and prayers are with the families."?

Coast Guard spokesman Carlos Vega told The Associated Press that the Coast Guard was turning over the remains to local authorities.

Earlier Saturday, the Coast Guard called off the search for the two workers. Three helicopter crews, a Coast Guard cutter and a fixed-wing aircraft crew had searched a 1,400-square-mile area around the platform, the Coast Guard said in a statement.

Coast Guard Capt. Peter Gautier said initial reports suggested that the explosion occurred when maintenance workers using a torch cut into a pipe with oil inside. Twenty-two people were on board the rig when the fire broke out and unleashed a black plume of smoke. Eleven workers were evacuated and nine others were taken by helicopter to hospitals.

Four workers airlifted to Louisiana's West Jefferson Medical Center suffered second- and third-degree burns to large parts of their bodies, said Taslin Alfonzo, a hospital spokeswoman.

The incident occurred a day after oil giant BP agreed to pay a record $4.5 billion in penalties for its role in the 2010 Gulf oil spill that killed 11 workers and spewed 4.9 million barrels of oil.

Since the Black Elk-operated rig was offline at the time of the fire, there was little risk of a major oil spill, officials said.

Eleven people were injured in the production platform blast and oil spillage was minimal, according to the Coast Guard. NBC's Brian Williams reports.

The platform sits in 56 feet of water some 17 miles south of Grand Isle, La., and production had been shut down since mid-August, Black Elk said.

The Bureau of Safety and Environmental Enforcement, which enforces offshore drilling regulations, is investigating the fire.

The fire was extinguished a few hours after the blast and Coast Guard Capt. Ed Cubanski told reporters that the platform appeared to be structurally sound. Twenty-two people had been aboard the rig at the time of the accident.

The platform is a shallow-water production platform, unlike?BP's Macondo well, which blew out in 2010?in mile-deep water.?The Macondo explosion killed 11 workers and caused the worst offshore oil spill in U.S. history.

The owner of the platform is Houston-based Black Elk Energy. On its website, the company stated that this month it was starting to drill the first of 23 new wells in the Gulf of Mexico.

Last Sunday,?The Houston Chronicle named Black Elk Energy?one of the top small businesses to work for in Houston based on employee surveys.

In August, the oil and gas company was named one of the fastest-growing privately held companies by Inc. Magazine.?

Probe defendant on Walker, GOP payroll

MILWAUKEE (AP) ? A court document says a former aide to Gov. Scott Walker who pleaded guilty to felony misconduct in office has been on the payroll of Walker?s campaign and the Wisconsin Republican Party.

Kelly Rindfleisch is to be sentenced Monday for doing campaign work from her office at the Milwaukee County Courthouse when she was an aide to Walker.
According to a sentencing report, Walker?s campaign gave her a job ?immediately after she left county service.?

A spokesman for the Walker campaign says Rindfleisch was hired by the state Republican Party to work on various administrative duties after the 2010 election.

Walker has said he is not a target of the John Doe investigation, but has cooperated with it.
The Journal Sentinel (http://bit.ly/T71oP7) reports Rindfleisch?s lawyer was unavailable for comment Friday.

Can the Burrowing Owl Population Rebound in North America?

Dear EarthTalk: What is the Burrowing Owl Conservation Network, and why is it so important to put so much effort into saving one species??Ginny Bateman, Portland, Ore.

Western burrowing owls (Athene cunicularia) are tiny, long-legged members of the owl family, native to the Americas and preferring open landscapes where they can dig new holes or use existing ones (such as abandoned prairie dog, skunk or armadillo homes) to nest and rear their young. Unlike most other owl species, these small but charismatic birds are more often seen out and about during daylight hours, but they are most active and do their primary feeding at night, preferring a diet of small rodents and large insects.

Once a common species in California and across North America, the Western burrowing owl?has become a rarer and rarer sight over the last three decades given habitat loss and other environmental perils the bird faces. Biologists consider the bird an indicator of wider ecosystem health, so if its population numbers are healthy then local ecosystems are likely thriving. But these days the bird is endangered in Canada and threatened in Mexico, is a state endangered species in Colorado, and is considered a ?species of special concern? in Florida and most of the western U.S. It is also listed on the International Union for Conservation of Nature?s (IUCN?s) international ?Red List? of endangered and threatened species (albeit as a species of ?Least Concern? in comparison with others in more dire straits).

A group of dedicated birders, conservationists, biologists and concerned citizens in San Francisco?s East Bay came together to protect dwindling burrowing owl populations locally but ended up creating a movement that spans the entire North American continent. Their group, the Burrowing Owl Conservation Network (BOCN) believes that burrowing owls are vital to maintaining healthy, functioning ecosystems, and as such is working on several fronts to help protect habitat and reverse the animal?s population slide.

First and foremost, BOCN works to conserve habitat for burrowing owls in California and beyond, including constructing artificial burrows to help re-establish burrowing owl colonies while larger ecosystem restoration efforts are underway. Teaching children and communities about ways they can help protect and improve burrowing owl habitat is another important part of BOCN?s work. Other ways the group helps the owls is by advocating for legislation and policy changes that encourage conservation of habitat, networking with like-minded individuals and institutions, and conducting field and laboratory research to increase understanding about how to help burrowing owls and create habitat conditions ideal for their survival.

The fact that burrowing owl populations in other parts of the world are rebounding gives hope to BOCN and other wildlife advocates and environmentalists. The birds are common and widespread in Central and South America, where they inhabit fields and even sometimes parks in urban areas.

Whether the birds can replicate their success in Latin America up north remains to be seen. As for what readers can do to help, learning about the behavior, biology and habitat needs of wild animals like the burrowing owl is a sure way to develop respect for nature?s inhabitants and a lifelong willingness to protect them. And nothing beats witnessing burrowing owls go about their rounds. Seeing them in the wild is enough to convince anyone that they are worth fighting to protect.
